VP23.09: Oxidative stress biomarkers in amniotic fluid in fetuses with isolated congenital heart defect

چکیده

Recent evidence suggests that oxidative stress (OS) plays an important role in the pathogenesis of congenital heart defect (CHD). The aim this study is to evaluate protein OS biomarkers amniotic fluid fetuses with isolated major CHD. This a retrospective including 92 samples, 50 cases diagnosis CHD and 42 controls (fetuses high risk genetic anomaly or extracardiac malformations, excluding CNS renal anomalies). All samples were collected from 2016 2020 at time performing after obtaining consent all patients. Five oxidation biomarkers, orto-tyrosine (o-tyr), metatyrosine (m-tyr), 3NO2-tyrosine (3NO2-tyr), 3I-tyrosine (3I-tyr), analysed provide preliminary results differences status between two groups, taking into account confusing factors such as gestational age (GA) amniocentesis condition maternal diabetes. Samples using Acquity-Xevo TQ system. No found groups terms GA (P = 0.8321) presence diabetes 0.182). After adjusting for GA, showed increased levels o-Tyr 0.038). other significant among rest analysed. Our data demonstrates than have biomarker fluid. Further studies evaluating greater number different its correlation postnatal outcome are warranted improve identification high-risk eventually new prevention strategies. Congenital heart defects and maternal biomarkers of oxidative stress.

BACKGROUND Women who have had pregnancies that were affected by nonsyndromic congenital heart defects have alterations in the homocysteine-methionine pathway that may indicate increased exposure to oxidative stress or reduced antioxidant defense or both. OBJECTIVE Our goal was to establish a maternal metabolic risk profile for nonsyndromic congenital heart defects that would enhance current p...

Elevated amniotic fluid amino acid levels in fetuses with gastroschisis.

Our objective was to measure maternal plasma and amniotic fluid amino acid concentrations in pregnant women diagnosed as having fetuses with gastroschisis in the second trimester of pregnancy. Twenty-one pregnant women who had fetuses with gastroschisis detected by ultrasonography (gastroschisis group) in the second trimester and 32 women who had abnormal triple screenings indicating an increas...

Identification of Symptomatic Fetuses Infected with Cytomegalovirus Using Amniotic Fluid Peptide Biomarkers

Cytomegalovirus (CMV) is the most common cause of congenital infection, and is a major cause of sensorineural hearing loss and neurological disabilities. Evaluating the risk for a CMV infected fetus to develop severe clinical symptoms after birth is crucial to provide appropriate guidance to pregnant women who might have to consider termination of pregnancy or experimental prenatal medical ther...

Functional genomic analysis of amniotic fluid cell-free mRNA suggests that oxidative stress is significant in Down syndrome fetuses.

To characterize the differences between second trimester Down syndrome (DS) and euploid fetuses, we used Affymetrix microarrays to compare gene expression in uncultured amniotic fluid supernatant samples. Functional pathway analysis highlighted the importance of oxidative stress, ion transport, and G protein signaling in the DS fetuses. Further evidence supporting these results was derived by c...

Hexosaminidase A in amniotic fluid of Tay-Sachs fetuses.

Hexosaminidase A is present in relatively low concentrations in cell-free amniotic fluids from pregnancies with Tay-Sachs fetuses. This isoenzyme was determined by an immunological procedure, radial immunodiffusion, by which hexosaminidase A can be directly and specifically detected, even in the presence of excess amounts of hexosaminidase B. No hexosaminidase A could be detected by the same pr...
